Selank is a synthetic heptapeptide derived from the endogenous tetrapeptide tuftsin, assigned CAS number 129954-34-3 and the development code TP-7. The molecule carries the formula C₃₃H₅₇N₁₁O₉ with a molecular weight of approximately 751.83 g/mol, and the full seven-residue sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. In purified, lab-tested form it is supplied as a white lyophilized powder. Selank was developed in the 1990s by the Institute of Molecular Genetics of the Russian Academy of Sciences in collaboration with the V. V. Zakusov State Institute of Pharmacology in Moscow, under the development code TP-7. The design team appended a C-terminal Pro-Gly-Pro tripeptide extension to the four-residue tuftsin core to block the standard sites of enzymatic cleavage, giving the analog substantially greater chemical stability than the parent compound while preserving the tuftsin pharmacophore. Among peptides of comparable size, Selank is frequently noted for its resistance to dipeptidyl peptidase cleavage at the C-terminal arginine and for its stability across aqueous buffer conditions encountered in standard characterization assays. Two consecutive proline residues following the C-terminal arginine create steric hindrance that resists serum aminopeptidases that would otherwise rapidly degrade unmodified tuftsin. Published preclinical research involving Selank — conducted entirely in laboratory and animal models — has examined the chemistry of stabilized tuftsin analogs and the structural principles that govern short-peptide proteolytic stability.
