GLP-1S is a synthetic 31-amino-acid peptide analogue of glucagon-like peptide-1 (GLP-1), supplied as a research chemical for in vitro and non-human laboratory study. Structurally it incorporates an alpha-aminoisobutyric acid (Aib) substitution at position 2 — which confers resistance to dipeptidyl peptidase-4 (DPP-4) cleavage — and a C18 fatty acid chain attached to Lys26 via a gamma-glutamyl-2x(OEG) spacer. The lipidation enables non-covalent binding to serum albumin in research matrices, a recognized methodology for extending circulating peptide half-life. In purified, lab-tested form it is supplied as a white lyophilized powder. GLP-1S was developed and characterized in the early 2010s as part of structure-activity research into long-acting GLP-1R agonists.
